Organocobalamin Reactions Relevant to the Mechanism of the a-Methyleneglutarate Mutase Enzyme [1]

چکیده
The organocobalamin with methylitaconic acid attached to cobalt via its methyl group has been synthesized for the first time. This compound is believed to be an intermediate in the interconver sion of methylitaconic acid and a-methyleneglutaric acid, catalysed by the coenzyme B 12-dependent a-m ethyleneglutarate mutase enzyme. Reactions of this organocobalamin and the corres ponding dimethyl ester demonstrate that cleavage of the Co —C bond leads to the rearranged amethyleneglutarate structure under conditions where intermediate carbanions are formed.

Concerning the intermediacy of organic radicals in vitamin B12-dependent enzymic reactions
in which the migrating group X can be OH, NH2 or a suitable substituted carbon atom such as C (=C H 2)C 02H. This paper discusses evidence for the participation of organic radicals as intermediates in these reactions. Theoretical and model studies supporting the intermediacy of radicals in the reactions catalysed by the enzymes diol dehydratase and a-methyleneglutarate mutase are described. For...
متن کاملConcerning the intermediacy of organic radicals in vitamin B12-dependent enzymic reactions.
The vitamin B12 coenzyme adenosylcobalamin assists the enzymic catalysis of molecular rearrangements of the type (formula; see text) in which the migrating group X can be OH, NH2 or a suitable substituted carbon atom such as C(=CH2)CO2H. This paper discusses evidence for the participation of organic radicals as intermediates in these reactions. Theoretical and model studies supporting the inter...
متن کاملRates of phosphorylation and dephosphorylation of phosphoglycerate mutase and bisphosphoglycerate synthase.
Phosphoglycerate mutase and bisphosphoglycerate synthase (mutase) can both be phosphorylated by either glycerate-1,3-P2 or glycerate-2,3-P2 to form phosphohistidine enzymes. The present study uses a rapid quench procedure to determine if, for each enzyme, the formation of the phosphorylated enzyme and phosphate transfer from the enzyme can occur at rates consistent with the overall reactions. W...
متن کاملEnzymes do what is expected (chalcone isomerase versus chorismate mutase).
Madicago sativa chalcone isomerase (CI) catalyzes the isomerization of chalcone to flavanone, whereas E. coli chorismate mutase (CM) catalyzes the pericyclic rearrangement of chorismate to prephenate. Covalent intermediates are not formed in either of the enzyme-catalyzed reactions, K(M) and k(cat) are virtually the same for both enzymes, and the rate constants (k(o)) for the noncatalyzed react...
متن کاملDistribution of two types of phosphoglyceric acid mutase, diphosphoglycerate mutase, and D-2,3-diphosphoglyceric acid.
the difference between the two types being in their requirement for 2,3-DPGA. Reactions 1 and 2 have been studied recently with crystalline and highly purified enzyme preparations (3-5). For clarity, in this paper the enzymes catalyzing these reactions are called 2,3DPGA dependent mutase and 2,3-DPGA independent mutase. We observed previously that the phosphoglyceric acid mu&se from several tis...
